EN 1657: 2016. Quantitative suspension test for the evaluation of fungicidal or yeasticidal activity of chemical disinfectants and antiseptics used in the veterinary area. Test method and requirements (phase 2, step 1).

Test accredited by ENAC (Spanish National Accreditation Entity), for standard mandatory microorganisms and additionals, including under conditions of the BPR guidance.

This test uses as mandatory organisms Candida albicans and Aspergillus brasiliensis for fungicidal activity and Candida albicans for levuricidal activity. Other fungi may optionally be used. The test must be performed at a mandatory temperature of 10°C and for a mandatory time of 30 minutes. Further, it may be performed with optional additional temperatures of 4, 20 or 40°C, and for optional additional times of 1, 5, 15 or 60 minutes. The test can be run in simulated conditions of low level dirty or in high level dirty conditions. For the yeasticidal activity of teat disinfection with Candida albicans, the mandatory test temperature is 30ºC and contact times of 30 seconds for pre-milking disinfection and 5 minutes for post-milking disinfection, with interfering substance from reconstituted skim milk. The product should be tested for at least three concentrations including one in the non-active range and one in the active range (these last two concentrations are selected by the testing laboratory). The product can be tested by the dilution-neutralization method, or by the membrane filtration method. To consider the fungicide product a log reduction of 4 in the number of viable cells must be demonstrated.
