Cosmetics testing - Microbiology of cosmetics. Detection of specified and non-specified microorganisms (EN ISO 18415: 2017).

The test provides information about the "presence or absence" of bacteria and yeast that must be absent in cosmetics: Staphylococcus aureus, Escherichia coli, Pseudomonas aeruginosa, Candida albicans) and other various bacteria in 1 g or 1 mL of the cosmetic sample. For test performing, the cosmetic is diluted in the ratio 1:10 in an enrichment broth directly, when products are miscible in water. When products are non-miscible in water are premixed with a solubilizing agent (polysorbate 80) and subsequently added to the enrichment broth. The enrichment broth should contain a neutralizer when tested product containing any antimicrobial agent as a preservative. Neutralizers used must have been previously validated by a validation test. The presence of microorganisms is determined by incubation for 24 to 72 hours at 32.5ºC in the enrichment broth, reseeding in selective and differential culture media to determine the presence of the afore mentioned specific microorganisms, and also in non-selective media to detect the presence of other than those microorganisms. The presence of other species of microorganisms is determined with appropriate identification tests. In IVAMI we perform identification of bacteria by sequencing of the 16S rRNA gene, and the yeast or filamentous fungi (molds) by sequencing the genomic region ITS-2.
