Corynebacterium diphtheriae - Neutralization test in guinea pig for detection and titration of antibodies against diphtheria toxin.

This test uses titrated diphtheria toxin to have the test dose that causes mild redness when introduced intradermally in guinea pig skin. Dilution of the toxin containing a test dose, faces several dilutions of the preparation of immunoglobulins, and after appropriate incubation mixtures test dose and dilution of immunoglobulin preparation is inoculated intradermally in the skin of guinea pigs. The animals are observed for 96 hours to know if there has been neutralized the effect of the toxin into guinea pig skin. According to the dilution immunoglobulin preparation that neutralize the effect of the toxin, antitoxic units existing against diphtheria toxin in the immunoglobulins product is determined.
